Cook studied physics and philosophy at Spelman College in Atlanta, and was heavily involved in student politics, once locking up the college's board of trustees to push them into divesting from apartheid-era South Africa. She later earned a further degree at Oxford in the UK, moving afterwards to Senegal to study the concept of time in African philosophy. But after wrestling with the distortions she witnessed in Dakar, she switched to economics.

One of her best-known papers, published in 2014, attempted to explain the divergence in economic outcomes between white individuals and African Americans in the US by looking at how lynchings of Black people after 1870 reduced the number of patents filed by Black inventors.
