There must be runners on first and second for that rule to be in effect. There is only a runner on first here.

Yes, always.
The rule aims to prevent the defense from turning a double or triple play by intentionally dropping the ball, deceiving the baserunners. The situation in the video only came about as the batter did not run to first. There is no need for the auto-out in this scenario if everyone is doing their jobs
